Using the router's DHCP and assigning the pi-hole as the routers DNS

Using the router's DHCP and assigning the pi-hole as DNS on each network device individually

Using the Pi-Hole DHCP and assigning the Router as DNS

Using the Pi-Hole DHCP and assigning a custom DNS

Any of the above. Once the client has an IP address it's going to be a point to point connection between the client and the DNS server. Any variation you see is going to be on the order of microseconds, and not perceptible to anyone.
